Upgrading tank using sand, water, LR, everything. Will the tank need to cycle?

Hey guys, this may sound like a silly question but if I upgrade in tank size using all of my LR, live sand, and water, obviously topping off the remainder with RO/DI water, will the tank need to cycle? Or will it already be considered a mature tank? Thanks!
